England are looking for their first win against the Springboks since 2006.
It is understood Tom Wood will start at open-side flanker, with Chris Robshaw and Billy Vunipola completing the back row.
Jonny May will also feature for the first time under boss Eddie Jones following his lengthy injury lay-off.
Forwards Charlie Ewels (Bath), Ellis Genge (Leicester), Teimana Harrison (Northampton) and Ben Morgan (Gloucester) return to their clubs.
As do backs Alex Goode (Saracens), Mike Haley (Sale Sharks), Alex Lozowski (Saracens), Semesa Rokoduguni (Bath) and Henry Slade (Exeter).
Jones will name his side to face South Africa on Thursday for the first of four games in their end-of-year series.
